چکیده

A spray-dried powder formulation obtained from the fermentation broth of the fungus has been developed. The effect of different additives (carriers and binders) on thermal deactivation of xylanase has been evaluated at 60°C. Addition of carriers and binders improves half-life by 6.6-7.0 folds, whereas the synergistic effect of carrier and binder enhances the half-life by 27.5 folds. Under the optimized conditions of the spray-drying process, retention of 99.4% specific activity is obtained with the maximum powder recovery of 62.4% at 140/65.2°C inlet/outlet temperature. The final spray dried powder produced is found to be brownish, free flowing and non-hygroscopic. The shelf-life of spray-dried powder is 202 days at 30°C, making it industrially useful product.

Thermal Stability and Thermodynamics of Xylanase from Melanocarpus albomyces in Presence of Polyols and Salts

An extracellular xylanase from the thermophilic fungus Melanocarpus albomyces IIS 68 was evaluated for its activity and stability in the presence of polyols and salts at 60 °C, and found to be an effective protecting agent for thermal deactivation of enzyme. Response surface methodology was employed to study the synergistic effects of glycerol and NaCl (thermostabilizers) for xylanase stability...

Investigate of Process Parameters on Xylanase Enzyme Activity in Melanocarpus Albomyces Batch Culture

The thermal and operational inactivation of a commercial Xylanase from Melanocarpus Albomyces was studied in several buffered solutions usually employed to study the activity of this enzyme. Some previous works have been done to understand the effect of the of the buffers sample on the enzyme activity. Afterwards, data obtained at temperatures from 40 to 80 C at pH 7.0 were used to find activit...

Myriocin, a new antifungal antibiotic from Myriococcum albomyces.

A new crystalline antifungal compound, myriocin, has been isolated from culture filtrates and mycelium of the thermophilic ascomycete Myriococcum albomyces. Myriocin is strongly active in vitro against yeasts and dermatophytes. This paper deals with the production, isolation, and purification, as well as the physical, chemical and biological properties of myriocin and one of its derivatives, an...

Bioprocess strategies for enhanced production of xylanase by Melanocarpus albomyces IITD3A on agro-residual extract.

The production of high titer xylanase without cellulase is required for prebleaching of pulps in pulp and paper industry. The mutant IITD3A of Melanocarpus albomyces developed from the spores of the wild type organism was used in this study. The statistical optimization of the process parameters by response surface methodology revealed that the production of xylanase was most affected by change...

XYLANASES OF MELANOCARPUS ALBOMYCES IIS 68 BIOSYNTHESIS, PURIFICATION AND CHARACTERIZATION BY VIBHOR SARASWAT Department of Biochemical Engineering and Biotechnology THESIS SUBMITTED FOR THE AWARD OF THE DEGREE OF DOCTOR OF PHILOSOPHY

Xylan is the major component of monocotyledon hemicelluloses of plant cell walls in which 13-1,4-linked D-xylose residues constitute the backbone of xylan. After cellulose, xylan is the most abundant renewable polysaccharide in nature, accounting for 20 to 30% of the dry weight of woody tissue. In order to maximize the hydrolysis of lignocellulosic residues by cellulase enzymes, synergistic act...
